Abstract
Objective—To report values for percentage coverage of the femoral head (PC) and Norberg angle (NA) in 4 common breeds of dogs and to determine values for each that distinguish between normal and dysplastic hip status on the basis of Orthopedic Foundation for Animals (OFA) hip evaluation.
Animals—1,841 dogs 24 to 48 months of age that were Labrador Retrievers (455), Golden Retrievers (423), Rottweilers (545), or German Shepherd Dogs (418).
Procedure—Retrospective analysis of NA and PC measured from standard OFA ventrodorsal pelvic radiographs from 4 breeds of dog.
Results—Norberg angle ranged from 67.4 to 124.4° for Labrador Retrievers, 59.7 to 128.6° for Rottweilers, 70.2 to 119.4° for Golden Retrievers, and 55.3 to 121.3° for German Shepherd Dogs. The PC ranged from 6.5 to 79.9% for Labrador Retrievers, 5.7 to 79.5% for Rottweilers, 8.3 to 79.3% for Golden Retrievers, and 5.4 to 83.7% for German Shepherd Dogs. On the basis of logistic regression modeling for determining normal versus abnormal hip status for all 4 breeds, cutoff points for NA were < 105° and PC were < 50%.
Conclusions and Clinical Relevance—Results of our study indicate that cutoff points of NA of 105° and PC of 50% do not differentiate normal versus dysplastic hip status. Each of the 4 breeds had different values for NA and PC that distinguished normal from dysplastic hip status. (Am J Vet Res 2000;61:1492–1500)
Abstract
Objective—To assess the prevalence and patterns of antimicrobial resistance among Escherichia coli strains isolated from the urine of women with cystitis or pyelonephritis and from fecal samples from dogs and healthy humans.
Design—Cross-sectional survey.
Sample Population—Escherichia coli isolates from 82 women with cystitis, 170 women with pyelonephritis, 45 dogs, and 76 healthy human volunteers.
Procedure—Susceptibility to 12 antimicrobial agents was determined by means of disk diffusion testing as specified by the NCCLS.
Results—Overall, the 4 most common antimicrobial resistance patterns were resistance to ampicillin, sulfisoxazole, trimethoprim, and trimethoprim-sulfamethoxazole (n = 45 [12% of all isolates]); ampicillin alone (33 [9%]); ampicillin and sulfisoxazole (29 [8%]); and sulfisoxazole alone (14 [4%]). None of the isolates were resistant to ceftazidime, ciprofloxacin, nitrofurantoin, or piperacillin-tazobactam. Resistance was significantly more common and extensive among isolates from women with cystitis or pyelonephritis than among isolates from healthy humans or dogs. Resistance was least common among isolates from dogs. The only resistance phenotype that was more common among canine isolates than human isolates was resistance to sulfisoxazole alone.
Conclusions and Clinical Relevance—Results suggest that dogs are unlikely to be an important external reservoir of antimicrobial-resistant E coli strains causing infections in humans. On the contrary, the data suggest that dogs conceivably could acquire resistant E coli strains from humans. ( J Am Vet Med Assoc 2004;225:368–373)
Abstract
Objective—To evaluate expression of cyclooxygenase (COX)-1 and COX-2 in the cornea, eyelid, and third eyelid of healthy horses and those affected with squamous cell carcinoma (SCC) by use of immunohistochemical techniques.
Animals—15 horses with SCC involving ocular tissues and 5 unaffected control horses.
Procedures—SCC-affected tissues were obtained from the cornea (n = 5 horses), eyelid (5), and third eyelid (5). Site-matched control tissues were obtained from 5 horses unaffected with SCC. Tissue sections of affected and control cornea, eyelid, and third eyelid were stained immunohistochemically for COX-1 and COX-2 via standard techniques. Stain uptake was quantified by use of computer-assisted image analysis of digital photomicrographs.
Results—Immunoreactivity for both COX-1 and COX-2 was significantly greater in equine corneas with SCC than in control corneas. No significant differences in COX-1 or COX-2 immunoreactivity were detected in eyelid and third-eyelid SCC, compared with site-matched control tissues.
Conclusions and Clinical Relevance—Immunoreactivity for COX-1 and COX-2 is high in equine corneal SCC, possibly indicating that COX plays a role in oncogenesis or progression of this tumor type at this site. Pharmacologic inhibition of COX may represent a useful adjunctive treatment for corneal SCC in horses.
Abstract
Objective—To determine immunoreactivity of matrix metalloproteinase (MMP)-1, -3, and -13 in cartilaginous tumors of dogs, correlate expression of MMP with histologic grade of tumors and clinical outcome of dogs, and compare MMP immunoreactivity between chondrosarcomas and chondromas.
Sample Population—Formalin-fixed, paraffin-embedded tissues obtained from samples of naturally occurring chondrosarcomas (n = 31) and chondromas (8) of dogs that were submitted to our veterinary medical diagnostic laboratory.
Procedure—Histologic sections from each sample were stained with H&E and monoclonal antibody to MMP-1, -3, and -13 by use of an avidin-peroxidase immunohistochemical technique. For each section, histologic grade (I, II, or III) and immunohistochemical expression (0, 1, 2, or 3) were evaluated. Clinical outcome was obtained from medical records or interviews with referring veterinarians and scored as a good outcome, moderate outcome, or poor outcome. Correlations among variables and differences between chondrosarcomas and chondromas were analyzed.
Results—Samples from chondrosarcomas had significantly higher immunoreactivity of MMP-1 and -13, compared with immunoreactivity in samples from chondromas. In chondrosarcomas, a significant positive correlation (r, 0.386) was found between MMP-1 and -13 immunoreactivities, and a significant negative correlation (r, –0.390) was detected between MMP-3 and -13 immunoreactivities.
Conclusion and Clinical Relevance—A significant increase in expression of collagenases (MMP-1 and - 13) in chondrosarcomas, compared with expression in chondromas, suggests that collagenases may play an important role in tumor progression, and possibly metastasis, in chondrosarcomas of dogs. Abstract
OBJECTIVE
To compare joint stability and ultimate strength among 4 prosthetic ligament constructs for repair of tarsal medial collateral ligament (MCL) injury in dogs.
SAMPLE
13 canine cadavers (26 hind limbs).
PROCEDURES
Each limb was stripped of all soft tissues except those associated with the tarsal joint and assigned to 1 of 4 prosthetic ligament constructs. The AN construct consisted of 3 bone anchors connected with monofilament nylon suture. The AU construct consisted of low-profile suture anchors connected with multifilament ultrahigh-molecular-weight polyethylene (UHMWPE) suture. The TN and TU constructs involved the creation of 3 bone tunnels and use of nylon or UHMWPE suture, respectively. Each limb underwent biomechanical testing before and after MCL transection and before and after cyclic range-of-motion testing following completion of the assigned construct. Tarsal joint stability (extent of laxity) was assessed with the joint in each of 3 positions (75°, 135°, and 165°). After completion of biomechanical testing, each limb was tested to failure to determine the ultimate strength of the construct.
RESULTS
Relative to intact tarsal joints, joint laxity was significantly increased following completion of all 4 constructs. Construct type was not associated with the magnitude of change in joint laxity. Ultimate strength was greatest for the UHMWPE-suture constructs.
CONCLUSIONS AND CLINICAL RELEVANCE
Results indicated that all 4 constructs effectively stabilized MCL-deficient tarsal joints. Implants used for the TU, TN, and AU constructs had a lower profile than those used for the AN construct, which may be clinically advantageous. In vivo studies are warranted.
Abstract
Objective—To characterize a 2007 bluetongue disease (BT) epizootic caused by bluetongue virus (BTV) serotype 17 in sheep in the Big Horn Basin of Wyoming.
Design—Cross-sectional study.
Animals—1,359 sheep from ranches in Wyoming and Montana.
Procedures—Information on clinical signs and history of BT in sheep was obtained from ranchers and attending veterinarians. At 3 to 6 months after the 2007 BT epizootic, blood samples were collected from rams, ewes, and lambs within and outside the Big Horn Basin; blood samples were also collected from lambs born in the spring of 2008. Sera were tested for anti-BTV antibodies by use of a competitive ELISA to determine the seroprevalence of BTV in sheep and to measure antibody titers. Virus isolation and reverse transcriptase PCR assays were used to determine long-term presence of the infectious virus or viral genetic material in RBCs of sheep.
Results—The percentage of sheep seropositive for BTV closely matched morbidity of sheep within flocks, indicating few subclinical infections. Flocks separated by as little as 1 mile had substantial variation in infection rate. Rams were infected at a higher rate than ewes. There was no evidence of BTV successfully overwintering in the area.
Conclusions and Clinical Relevance—This epizootic appears to be a new intrusion of BTV into a naïve population of sheep previously protected geographically by the mountains surrounding the Big Horn Basin. Rams may have a higher infection rate as a result of increased vector biting opportunity because of the large surface area of the scrotum.
Abstract
Objective—To determine clinical and radiographic findings, treatment, and outcome for llamas with long-bone fractures.
Design—Retrospective study.
Animals—6 llamas.
Procedure—Medical records of llamas admitted between 1993 and 1998 because of long-bone fractures were reviewed. Data collected included age, sex, type of fracture, method of fracture repair, and postoperative complications. The Fisher exact test was used to compare age and sex of the llamas with long-bone fractures with those of the hospital population of llamas. All owners were contacted by telephone to determine perceived postoperative problems and whether the llamas were able to perform as expected.
Results—Mean age was 160.8 days (range, 23 to 365 days). There was 1 male and 5 females. Fractures were more likely to occur in young llamas (≤ 1 year old) than in adults. Five of the fractures were attributed to traumatic episodes. Long bones affected included the tibia (n = 2), radius (2), femur (1), and humerus (1). Internal fixation with lag screws, plating, or both was performed on fractures of all llamas except 1; that llama was treated by use of confinement to a stall. None of the llamas had intraoperative complications, but postoperative complications were reported in 2 llamas. All fractures healed eventually, and clients were pleased with outcomes.
Conclusions and Clinical Relevance—Long-bone fractures in llamas are uncommon. Several types of long bone fractures can be successfully repaired by use of internal fixation, resulting in few complications and minimal convalescent time. (J Am Vet Med Assoc 2000;216:1291–1293)
Abstract
Objective—To determine the expression and distribution of a disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), its substrates aggrecan and versican, and their binding partner hyaluronan in laminae of healthy horses.
Sample—Laminae from the forelimb hooves of 8 healthy horses.
Procedures—Real-time quantitative PCR assay was used for gene expression analysis. Hyaluronidase, chondroitinase, and keratanase digestion of lamina extracts combined with SDS-PAGE and western blotting were used for protein and proteoglycan analysis. Immunofluorescent and immunohistochemical staining of tissue sections were used for protein and hyaluronan localization.
Results—Genes encoding ADAMTS-4, aggrecan, versican, and hyaluronan synthase II were expressed in laminae. The ADAMTS-4 was predominantly evident as a 51-kDa protein bearing a catalytic site neoepitope indicative of active enzyme and in situ activity, which was confirmed by the presence of aggrecan and versican fragments bearing ADAMTS-4 cleavage neoepitopes in laminar protein extracts. Aggrecan, versican, and hyaluronan were localized to basal epithelial cells within the secondary epidermal laminae. The ADAMTS-4 localized to these cells but was also present in some cells in the dermal laminae.
Conclusions and Clinical Relevance—Within digital laminae, versican exclusively and aggrecan primarily localized within basal epithelial cells and both were constitutively cleaved by ADAMTS-4, which therefore contributed to their turnover. On the basis of known properties of these proteoglycans, it is possible that they can protect the basal epithelial cells of horses from biomechanical and concussive stress.
Abstract
Objective—To determine whether increased gene expression of a disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4) in laminae of horses with starch gruel–induced laminitis was accompanied by increased enzyme activity and substrate degradation.
Sample—Laminae from the forelimb hooves of 8 healthy horses and 17 horses with starch gruel–induced laminitis (6 at onset of fever, 6 at onset of Obel grade 1 lameness, and 5 at onset of Obel grade 3 lameness).
Procedures—Gene expression was determined by use of cDNA and real-time quantitative PCR assay. Protein expression and processing were determined via SDS-PAGE and quantitative western blotting. Protein distribution and abundance were determined via quantitative immunofluorescent staining.
Results—ADAMTS-4 gene expression was increased and that of versican decreased in laminitic laminae, compared with expression in healthy laminae. Catalytically active ADAMTS-4 also was increased in the tissue, as were ADAMTS-4–cleavage fragments of versican. Immunofluorescent analyses indicated that versican was depleted from the basal epithelia of laminae of horses at onset of Obel grade 3 lameness, compared with results for healthy laminae, and this was accompanied by regional separation of basal epithelial cells from the basement membrane. Aggrecan gene and protein expression were not significantly affected.
Conclusions and Clinical Relevance—Changes in gene and protein expression of ADAMTS-4 and versican in the basal epithelium of laminitic laminae indicated a fundamental change in the physiology of basal epithelial cells. This was accompanied by and may have caused detachment of these cells from the basement membrane.
